✔️ Vave is used to relieve:

• Dyspeptic symptoms associated with delayed gastric emptying, GERD, and esophagitis:
• Epigastric fullness, bloating, upper abdominal pain
• Belching, flatulence, and early satiety
• Nausea, vomiting, heartburn (with/without regurgitation)
• Non-ulcer dyspepsia
• Acute nausea and vomiting (functional, organic, infectious, diet-related, post-radiotherapy, drug-induced, or migraine-related)
• Parkinson’s disease: management of dopamine-agonist-induced nausea and vomiting
• Radiological studies: enhancing barium transit in follow-through procedures

✔️ Side Effects

• Rare: mild intestinal cramps
• Rare extrapyramidal reactions (mostly in young children)
• Hyperprolactinemia → galactorrhea, gynecomastia (rare)
• Possible allergic reactions (rash, urticaria)

✔️ Dosage & Administration

Adults: 10–20 mg (tablet/suspension) every 6–8 hours (max 80 mg/day)

Children: 0.2–0.4 mg/kg every 6–8 hours (max treatment: 12 weeks)

Take 15–30 minutes before meals; may also be taken before bedtime if needed

✔️ Drug Interactions

Anticholinergics reduce antidyspeptic effects

Antacids/antisecretory agents lower bioavailability

CYP3A4 inhibitors (azole antifungals, macrolides, HIV protease inhibitors, and nefazodone) may raise plasma levels

Enhances tolerance of dopaminergic agonists (L-DOPA, bromocriptine) by reducing peripheral side effects

✔️ Contraindications

• Hypersensitivity to domperidone
• Neonates
• GI hemorrhage, obstruction, or perforation
• Prolactin-releasing pituitary tumor (prolactinoma)

✔️ Pregnancy & Lactation

No proven teratogenicity, but use in the 1st trimester only if clearly indicated

Excreted in breast milk (low levels) → breastfeeding generally not recommended unless benefits outweigh risks

✔️ Precautions & Warnings

• Use with caution in children (risk of extrapyramidal effects due to an immature blood-brain barrier)
• Caution in hepatic impairment (extensively metabolized in the liver)
• In renal impairment, reduce dosing frequency for long-term use
• Not recommended in neonates
• Monitor long-term therapy carefully
