✔️ Side Effects

Rhabdomyolysis, myopathy, liver enzyme abnormalities, myalgia, abdominal pain, increased hepatic enzymes.

✔️ Dosage & Administration:

• Starting Dosage: 10/10 mg or 20/10 mg daily, which can be taken as a single dose at any time of the day, with or without food.
• For patients needing a larger LDL-C reduction (>55%), the starting dose is 40/10 mg daily.
• Homozygous Familial Hypercholesterolemia: The recommended dosage is 40/10 mg or 80/10 mg daily.
• Lipid levels should be checked 2 or more weeks after initiation or dose adjustments, and dosage should be adjusted as needed.

• Pediatric Use: Not established in pediatric patients.
• Geriatric Use: No dosage adjustment needed for elderly patients.

✔️ Interactions:

• Increased risk of myopathy when combined with fibric acid derivatives, niacin, cyclosporine, or strong CYP3A4 inhibitors (e.g., clarithromycin, HIV protease inhibitors, itraconazole).
• Gemfibrozil: Avoid concurrent use due to an increased risk of rhabdomyolysis.
• Warfarin: INR should be monitored if used with warfarin.
• Colchicine: Caution should be exercised when coadministering with colchicine.

• Active liver disease or persistent unexplained elevations in hepatic transaminase levels.
• Hypersensitivity to any components of the preparation

✔️ Pregnancy & Lactation:

• Pregnancy Category X: Tiginor EZ is contraindicated in pregnant women or those who may become pregnant. It is not recommended during pregnancy because lipid-lowering drugs do not benefit the fetus and can interfere with fetal development.
• It is unknown if atorvastatin is excreted in human milk, but similar drugs may pass into breast milk.

✔️ Precautions & Warnings:

• Myopathy: Be cautious with colchicine coadministration due to an increased risk of rhabdomyolysis.
• Liver Enzymes: Liver function tests should be conducted before starting treatment and monitored as needed. Discontinue if liver injury occurs.
• Endocrine Function: Caution is advised when combined with drugs that may affect steroid hormone levels.
