Voriconazole is recommended for the treatment of the following fungal infections in individuals aged 12 and up:

1. Invasive Aspergillosis
2. Candidemia (non-tuberculous) and widespread candidiasis in the skin, abdomen, kidneys, bladder wall, and wounds
3. Candida esophagitis
4. Infections caused by Scedosporium apiospermum and Fusarium species, especially Fusarium solani, in patients resistant to or intolerant of alternative treatments.

✔️ Pharmacology

Voriconazole is a triazole antifungal drug that inhibits CYP450-dependent 14-alpha sterol demethylase, binding to and inhibiting ergosterol production. Inhibition of 14-alpha sterol demethylase leads to ergosterol depletion in fungal cell membranes.

✔️ Dosage & Administration:

Oral:

• Loading dose regimen (for the first 24 hours):
  • At or over 40 kg body weight: 400 mg or 10 ml every 12 hours
  • Below 40 kg body weight: 200 mg or 5 ml every 12 hours
• Maintenance dose (after first 24 hours):
  • At or over 40 kg body weight: 200 mg or 5 ml twice daily
  • Below 40 kg body weight: 100 mg or 2.5 ml twice daily

• Voriconazole interacts with CYP3A4, CYP2C9, and CYP2C19 inhibitors and inducers. Dosage adjustments are necessary.
